Extraction of DNA from Plant Tissues

Objectives

To demonstrate extraction of DNA and to study the characteristics of plant DNA.

Introduction

Deoxyribonucleic Acid (DNA) is a genetic material that carries biological data for the formation and function of living things. Within the plant species, the DNA molecule houses structures of nucleus, mitochondria and chloroplasts; which the chloroplasts makes plant DNA unique from animal DNA. In a simple DNA molecule, four nucleotides can be found. For each nucleotide it houses five-carbon sugar (deoxyribose), phosphate and one of four nitrogenous bases; which either one could be adenine (A) and guanine (G) the double-ringed purines or thymine (T) and cytosine (C) the single-ringed pyrimidines, as explained by (International Baccalaureate Biology, 2013).

The hypothesis in the experiment is that the shampoo will be able to break through the phospholipid bilayer of the strawberry plant cell and DNA will be precipitated when ethanol is added.

Method

3g of salt, 90ml of distilled water and 10ml of hair shampoo were added together in the 100ml beaker while stirring.

Chopped strawberries were adding into mixture after it is stirred thoroughly.

Beaker of mixture was given an exact 15 minutes of 60ºC water bath.

After 15 minutes of hot water bath, beaker of mixture was given an ice water bath.

Stirring of the mixture was until temperature of the beaker and mixture cooled down.

Mixture was filled into a blender to blender everything thoroughly (Figure 1.1).
